Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_004364.5(CEBPA):c.310_315dup (p.Gly104_Asp105dup), citing Ambry Variant Classification Scheme 2023. This variant lies in the CEBPA gene (transcript NM_004364.5) at coding-DNA position 310 through coding-DNA position 315, duplicating 6 bases. Submitter rationale: The c.310_315dupGGCGAC variant (also known as p.G104_D105dup), located in coding exon 1 of the CEBPA gene, results from an in-frame duplication of GGCGAC at nucleotide positions 310 to 315. This results in the duplication of 2 extra residues (GD) between codons 104 and 105. These amino acid positions are well conserved in available vertebrate species. In addition, this variant is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
