Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 3' splice acceptor site. This is expected to result in a frameshift or skipping of the last exon of the TMEM237 gene. However, these predictions have yet to be confirmed by functional studies. To our knowledge truncations in the last exon of TMEM267 gene have not been reported. The variant allele was found at a frequency of 4.1e-06 in 245018 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1160-1G>A in individuals affected with Joubert Syndrome And Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
