Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000601.6(HGF):c.1602G>C (p.Gln534His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with histidine, which is basic and polar, at codon 534 of the HGF protein (p.Gln534His). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with HGF-related conditions. ClinVar contains an entry for this variant (Variation ID: 1472755). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change does not substantially affect HGF function (PMID: 1321034, 1661588). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
