NM_002292.4(LAMB2):c.2578C>T (p.Arg860Cys) was classified as Uncertain significance for LAMB2-related infantile-onset nephrotic syndrome; Pierson syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces arginine with cysteine at codon 860 of the LAMB2 protein (p.Arg860Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is present in population databases (rs563405094, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with LAMB2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
