Likely pathogenic for Spondyloepimetaphyseal dysplasia with joint laxity; Ehlers-Danlos syndrome, spondylodysplastic type, 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_080605.4(B3GALT6):c.530GCC[5] (p.Arg180dup), citing Invitae Variant Classification Sherloc (09022015): This variant, c.539_541dup, results in the insertion of 1 amino acid(s) of the B3GALT6 protein (p.Arg180dup), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with clinical features of spondyloepimetaphyseal dysplasia (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
