NM_001903.5(CTNNA1):c.176A>G (p.His59Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H59R variant (also known as c.176A>G), located in coding exon 2 of the CTNNA1 gene, results from an A to G substitution at nucleotide position 176. The histidine at codon 59 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
