Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004836.7(EIF2AK3):c.1507A>G (p.Asn503Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with aspartic acid at codon 503 of the EIF2AK3 protein (p.Asn503Asp). The asparagine residue is weakly conserved and there is a small physicochemical difference between asparagine and aspartic acid. This variant is present in population databases (rs768630246, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with EIF2AK3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
