NM_024426.6(WT1):c.785-14G>A was classified as Uncertain Significance for Wilms tumor 1 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant causes a G>A nucleotide substitution at the -14 position of intron 2 of the WT1 gene. Splice site prediction tools are inconclusive regarding the impact of this variant on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with WT1-related disorders in the literature. This variant has been identified in 5/258694 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
