Uncertain significance for Charcot-Marie-Tooth disease axonal type 2O — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001376.5(DYNC1H1):c.10174A>G (p.Met3392Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 3392 of the DYNC1H1 protein (p.Met3392Val). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and valine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with epileptic encephalopathy (PMID: 28325891). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt DYNC1H1 protein function.
