Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000238.4(KCNH2):c.64T>G (p.Phe22Val), citing Ambry Variant Classification Scheme 2023: The p.F22V variant (also known as c.64T>G), located in coding exon 1 of the KCNH2 gene, results from a T to G substitution at nucleotide position 64. The phenylalanine at codon 22 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
