Submitter rationale: The p.R508K variant (also known as c.1523G>A), located in coding exon 16 of the EYA4 gene, results from a G to A substitution at nucleotide position 1523. The arginine at codon 508 is replaced by lysine, an amino acid with highly similar properties. This alteration has been reported in a biobank cohort (Ahmadmehrabi S et al. Hum Genet, 2021 Jun;140:957-967). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
