Uncertain significance for Mitochondrial DNA depletion syndrome 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003849.4(SUCLG1):c.113T>C (p.Ile38Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with threonine at codon 38 of the SUCLG1 protein (p.Ile38Thr). The isoleucine residue is weakly conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SUCLG1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
