This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 1390, where G is replaced by A; at the protein level this means replaces glycine at residue 464 with serine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This sequence change replaces glycine with serine at codon 464 of the SDHA protein (p.Gly464Ser). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SDHA-related conditions.
