Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017654.4(SAMD9):c.2704C>T (p.Arg902Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 902 of the SAMD9 protein (p.Arg902Trp). This variant is present in population databases (rs763861072, gnomAD 0.003%). This missense change has been observed in individual(s) with clinical features of SAMD9-related conditions (PMID: 34621053). ClinVar contains an entry for this variant (Variation ID: 1468906). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on SAMD9 protein function. Experimental studies have shown that this missense change affects SAMD9 function (PMID: 34621053).
