Uncertain significance for Schimke immuno-osseous dysplasia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014140.4(SMARCAL1):c.709C>A (p.Gln237Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with lysine at codon 237 of the SMARCAL1 protein (p.Gln237Lys). The glutamine residue is weakly conserved and there is a small physicochemical difference between glutamine and lysine. This variant is present in population databases (rs754790488, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with SMARCAL1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
