NM_006231.4(POLE):c.5732G>C (p.Trp1911Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W1911S variant (also known as c.5732G>C), located in coding exon 42 of the POLE gene, results from a G to C substitution at nucleotide position 5732. The tryptophan at codon 1911 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
