Uncertain significance for Dyskeratosis congenita — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001099274.3(TINF2):c.1211G>A (p.Gly404Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with glutamic acid at codon 404 of the TINF2 protein (p.Gly404Glu). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and glutamic acid. This variant is present in population databases (rs765918306, ExAC 0.001%). This variant has not been reported in the literature in individuals with TINF2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
