Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.2585C>G (p.Ala862Gly), citing Ambry Variant Classification Scheme 2023: The p.A862G variant (also known as c.2585C>G), located in coding exon 22 of the TSC2 gene, results from a C to G substitution at nucleotide position 2585. The alanine at codon 862 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
