NM_004104.5(FASN):c.2078C>T (p.Pro693Leu) was classified as Uncertain significance for Epileptic encephalopathy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with FASN-related conditions. This variant is present in population databases (rs764572985, ExAC 0.002%). This sequence change replaces proline with leucine at codon 693 of the FASN protein (p.Pro693Leu). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and leucine.
