Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000190.4(HMBS):c.65G>A (p.Arg22His), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HMBS c.65G>A (p.Arg22His) results in a non-conservative amino acid change located in the porphobilinogen deaminase, N-terminal domain (IPR022417) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4.4e-05 in 251404 control chromosomes (gnomAD). To our knowledge, no occurrence of c.65G>A in individuals affected with Acute Intermittent Porphyria and no experimental evidence demonstrating its impact on protein function have been reported. One submitter has cited a clinical-significance assessment for this variant to ClinVar after 2014 and has classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
