This variant lies in the SMARCA4 gene (transcript NM_003072.5) at coding-DNA position 4252, where G is replaced by T; at the protein level this means replaces aspartic acid at residue 1418 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with tyrosine at codon 1450 of the SMARCA4 protein (p.Asp1450Tyr). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and tyrosine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
