This variant has not been reported in the literature in individuals affected with HEXA-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 37 of the HEXA protein (p.Tyr37Cys). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant disrupts the p.Tyr37Asn amino acid residue in HEXA. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 18358410, 33083013). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
