NM_000548.5(TSC2):c.3812C>G (p.Thr1271Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T1271R variant (also known as c.3812C>G), located in coding exon 30 of the TSC2 gene, results from a C to G substitution at nucleotide position 3812. The threonine at codon 1271 is replaced by arginine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
