Uncertain significance for Familial hypocalciuric hypercalcemia; Autosomal dominant hypocalcemia 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000388.4(CASR):c.1054T>C (p.Trp352Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with arginine at codon 352 of the CASR protein (p.Trp352Arg). The tryptophan residue is highly conserved and there is a moderate physicochemical difference between tryptophan and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with CASR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
