This variant lies in the IRF6 gene (transcript NM_006147.4) at coding-DNA position 1400, where A is replaced by G; at the protein level this means replaces glutamine at residue 467 with arginine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with IRF6-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
