Uncertain significance for Progressive microcephaly-seizures-cortical blindness-developmental delay syndrome; Autosomal dominant nonsyndromic hearing loss 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005219.5(DIAPH1):c.3463C>G (p.Arg1155Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DIAPH1 gene (transcript NM_005219.5) at coding-DNA position 3463, where C is replaced by G; at the protein level this means replaces arginine at residue 1155 with glycine — a missense variant. Submitter rationale: This sequence change replaces arginine with glycine at codon 1155 of the DIAPH1 protein (p.Arg1155Gly). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and glycine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DIAPH1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
