Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001297.5(CNGB1):c.1393C>T (p.Pro465Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with serine at codon 465 of the CNGB1 protein (p.Pro465Ser). The proline residue is moderately conserved and there is a moderate physicochemical difference between proline and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with CNGB1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CNGB1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
