Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.1607C>A (p.Thr536Asn), citing Ambry Variant Classification Scheme 2023: The p.T536N variant (also known as c.1607C>A), located in coding exon 10 of the RAD50 gene, results from a C to A substitution at nucleotide position 1607. The threonine at codon 536 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
