NM_021629.4(GNB4):c.311C>T (p.Ala104Val) was classified as Uncertain significance for Charcot-Marie-Tooth disease dominant intermediate F by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs761510333, ExAC 0.002%). This sequence change replaces alanine with valine at codon 104 of the GNB4 protein (p.Ala104Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant has not been reported in the literature in individuals with GNB4-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
