NM_007294.4(BRCA1):c.2135G>A (p.Cys712Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C712Y variant (also known as c.2135G>A), located in coding exon 9 of the BRCA1 gene, results from a G to A substitution at nucleotide position 2135. The cysteine at codon 712 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
