Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002317.7(LOX):c.1030A>G (p.Thr344Ala), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available"). This variant has not been reported in the literature in individuals affected with LOX-related conditions. This variant is present in population databases (rs535032095, ExAC 0.02%), and has an allele count higher than expected for a pathogenic variant (PMID: 28166811). This sequence change replaces threonine with alanine at codon 344 of the LOX protein (p.Thr344Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine.
