Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018993.4(RIN2):c.-36-2787C>T, citing Invitae Variant Classification Sherloc (09022015): The RIN2 gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_001242581.1, and corresponds to NM_018993.3:c.-2823C>T in the primary transcript. This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 37 of the RIN2 protein (p.Thr37Met). This variant is present in population databases (rs776636138, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RIN2-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
