Uncertain significance for Marfan syndrome; Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000138.5(FBN1):c.7799A>G (p.Tyr2600Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 7799, where A is replaced by G; at the protein level this means replaces tyrosine at residue 2600 with cysteine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt FBN1 protein function. This variant has not been reported in the literature in individuals with FBN1-related conditions. This sequence change replaces tyrosine with cysteine at codon 2600 of the FBN1 protein (p.Tyr2600Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency).
