Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.299G>A (p.Cys100Tyr), citing Ambry Variant Classification Scheme 2023: The p.C100Y variant (also known as c.299G>A), located in coding exon 1 of the HOXB13 gene, results from a G to A substitution at nucleotide position 299. The cysteine at codon 100 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
