NM_006516.4(SLC2A1):c.142T>G (p.Trp48Gly) was classified as Uncertain significance for GLUT1 deficiency syndrome 1, autosomal recessive by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with SLC2A1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces tryptophan with glycine at codon 48 of the SLC2A1 protein (p.Trp48Gly). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and glycine. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SLC2A1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
