Uncertain significance for Peroxisome biogenesis disorder 9B — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000288.4(PEX7):c.330C>G (p.His110Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with glutamine at codon 110 of the PEX7 protein (p.His110Gln). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and glutamine. This variant is present in population databases (rs199648976, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with PEX7-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
