Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001171.6(ABCC6):c.2302T>C (p.Tyr768His), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ABCC6 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with ABCC6-related conditions. This sequence change replaces tyrosine with histidine at codon 768 of the ABCC6 protein (p.Tyr768His). The tyrosine residue is highly conserved and there is a moderate physicochemical difference between tyrosine and histidine. This variant is not present in population databases (ExAC no frequency).
