Pathogenic for Exostoses, multiple, type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000011.9:g.(?_44129235)_(44129808_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Deletion of exon 2 ‚Äãhas been observed in individual(s) with multiple osteochondromas (PMID: 15221792, 21703028) This variant is a gross deletion of the genomic region encompassing exon(s) 2 of the EXT2 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 2 of the EXT2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in EXT2 are known to be pathogenic (PMID: 10679937, 19810120).
