Pathogenic for Cerebral cavernous malformation 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.11:g.(?_167413364)_(167437945_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with PDCD10-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 2-5 of the PDCD10 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 5 of the PDCD10 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in PDCD10 are known to be pathogenic (PMID: 15543491, 18300272, 23801932). For these reasons, this variant has been classified as Pathogenic.
