Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001163435.3(TBCK):c.2479G>T (p.Glu827Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu827*) in the TBCK gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in TBCK are known to be pathogenic (PMID: 27040692, 30103036). This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with TBCK-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
