Pathogenic for Heterotaxy, visceral, 5, autosomal — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000010.10:g.(?_72183370)_(72201347_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of exons 2-3 and part of exon 1 (c.77_*9322del) of the NODAL gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with NODAL-related conditions. This variant deletes the mature protein domain (p.His238-Leu347) of the NODAL protein that is required for downstream signaling and proper embryonic development (PMID: 12447384, 18579681). For these reasons, this variant has been classified as Pathogenic.
