NC_000022.10:g.(?_41553153)_(41554524_?)del was classified as Pathogenic for Rubinstein-Taybi syndrome due to EP300 haploinsufficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with EP300-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 18-19 of the EP300 gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Loss-of-function variants in EP300 are known to be pathogenic (PMID: 15706485, 24476420).
