NC_000002.11:g.(?_20153575)_(20153759_?)del was classified as Pathogenic for Cranioectodermal dysplasia 2; Short-rib thoracic dysplasia 7 with or without polydactyly by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant has not been reported in the literature in individuals with WDR35-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 13 of the WDR35 gene. This deletion is out-of-frame, and is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Loss-of-function variants in WDR35 are known to be pathogenic (PMID: 22486404, 29068549).
