This variant lies in the COL7A1 gene (transcript NM_000094.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 6751 through the canonical splice acceptor site of the intron immediately before coding-DNA position 6751, deleting this region. Submitter rationale: Variant summary: COL7A1 c.6751-2_6751-1delAG is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of COL7A1 function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 251150 control chromosomes. c.6751-2_6751-1delAG has been reported in the literature in multiple individuals affected with Dystrophic Epidermolysis Bullosa, Recessive (example, Jerabkova _2010, Kopeckova_2016) . These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
