This variant has been observed in individual(s) with clinical features of TUBB2A-related conditions (Invitae). In at least one individual the variant was observed to be de novo. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change replaces alanine with aspartic acid at codon 314 of the TUBB2A protein (p.Ala314Asp). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and aspartic acid.
