Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000268.4(NF2):c.1612C>T (p.Gln538Ter), citing Ambry Variant Classification Scheme 2023: The p.Q538* pathogenic mutation (also known as c.1612C>T), located in coding exon 15 of the NF2 gene, results from a C to T substitution at nucleotide position 1612. This changes the amino acid from a glutamine to a stop codon within coding exon 15. This mutation has been detected in an individual with NF2-related schwannomatosis (Bourn D et al. Hum Genet, 1995 May;95:572-4). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
