NC_000014.8:g.(?_73673074)_(73673200_?)del was classified as Pathogenic for Alzheimer disease 3; Pick disease; Acne inversa, familial, 3; Frontotemporal dementia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that deletion of exon 9 affects PSEN1 protein function (PMID: 16959576). Deletions of exon 9 have been observed in individual(s) with early-onset Alzheimer's disease (PMID: 10502791, 11198283, 10854108). It has also been observed to segregate with disease in related individuals. This variant is a gross deletion of the genomic region encompassing exon(s) 9 of the PSEN1 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame.
