Pathogenic for Congenital myasthenic syndrome 10; Fetal akinesia deformation sequence 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000004.11:g.(?_3465103)_(3465298_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 1-2 of the DOK7 gene, which includes the initiator codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DOK7 are known to be pathogenic (PMID: 16794080, 16917026, 18626973, 19261599). A similar copy number variant has been observed in individual(s) with congenital myasthenic syndrome (PMID: 28508085). For these reasons, this variant has been classified as Pathogenic.
