This variant lies in the EXT1 gene (transcript NM_000127.3) at coding-DNA position 837, duplicating one base; at the protein level this means shifts the reading frame starting at arginine residue 280, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has been observed in individual(s) with multiple osteochondromas (PMID: 21499719). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Arg280Glnfs*9) in the EXT1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in EXT1 are known to be pathogenic (PMID: 10679937, 11391482, 19810120).
