This variant disrupts the C-terminus of the COL6A1 protein. Other variant(s) that disrupt this region (p.Ala778Profs*53) have been determined to be pathogenic (PMID: 23738969). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. This variant has not been reported in the literature in individuals with COL6A1-related conditions. This variant results in the deletion of exons 33-35 and part of exon 32 (c.2225_*54del) of the COL6A1 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product.
